Then special treatment begins: Pretreatment Agarose is used for embedding bacteria on the filter, so it's not possible wash bacteria from the filter while using different solutions through the process. Enzymatic pretreatment is used to disrupt bacterial cell walls and prepare them being accessible for probes during hybridization. Method Fluorescence In Situ Hybridization visualizes selected group/lineage among the whole prokaryote population. Its principle is to combine a DNA probe with a complementary region in the ribosome. While FISH probes have attached a fluorescent dye, CARD-FISH probes have the horseradish peroxidase enzyme and the fluorochrome is deposited by its activity, which results in an amplified signal and enables to differentiate small or low active cells.
